What did people taking retatrutide notice first?

The interview study was small and relied on what people remembered. It spoke with 40 people from a Phase 2 trial [10]. Among the 36 who received retatrutide, 31 noticed eating changes within 8 weeks.

They described less hunger, fullness after less food, smaller helpings, and different food choices. They also felt more in charge of eating. These were memories from study members, not readings from a machine.

Hunger can change before the scale moves much. Your weight reflects many days of eating and using fuel, so it changes more slowly. GLP-1, a gut hormone that acts in the brain, may help explain why thoughts of food grew quiet [7].

How did weight and blood sugar change in weeks 8-24?

The main 48-week Phase 2 obesity study checked weight several times [1]. Early on, all four retatrutide groups—1, 4, 8, and 12 mg—had a larger average loss than people given placebo. By week 48, placebo weight had changed about −2.1%. At week 48, averages for the drug groups were still falling.

The opening study lasted only 12 weeks [4]. It gave an early 12-week view of people with type 2 diabetes before the larger Phase 2 work. At the largest amount, average loss was 8.96 kilograms greater than with placebo. The authors were 90% sure the true average gap fell within their stated span, but its exact size remains uncertain.

In the diabetes Phase 2 study, the three-month blood sugar test fell −2.02 points with 12 mg after 24 weeks [2]. A change by week 24 means blood sugar had started improving earlier. The test looks back over time and doesn't show every daily high or low.

What kept changing between weeks 24-48?

By week 48, the largest-amount groups still hadn't reached a flat weight line [1]. At week 48, average loss with 12 mg was −24.2%. Phase 2, the mid-sized test, can't tell whether weight would later level off, keep falling, or return.

The Phase 2 liver result also lasted. Liver fat fell −82.4% with 12 mg by week 24 and −86.0% with 12 mg by week 48 [5]. That is all the short study can show.

One review from 2025 covered Phase 1/2 work, meaning the first and mid-sized studies, and offered two possible reasons [6]. GLP-1 may reduce hunger, while glucagon may draw more heavily on stored fuel. Phase 3 is the larger test of that idea, since the reason for the difference from older drugs hasn't been proved.

What is known after a person stops retatrutide?

The length of any benefit after retatrutide stops is unknown. By mid-2026, no published Phase 2 study had followed people after treatment ended. With similar drugs based on a gut hormone, people often regain much of the weight.

Glucagon may increase fuel use during treatment. No study has shown that this effect lasts after the drug is gone. No finished retatrutide study has watched people after stopping for enough time to tell you whether the weight stays off.
